Lipophilicity of drug like molecules with HPLC



Abstract Lipophilicity has been shown to be of paramount importance in several ADME aspects, that is absorption, distribution, metabolism and excretion. It is generally held that very lipophilic compounds are “preferred” targets for metabolism, often leading to high clearance values and, quite often, lipophilicitity positively correlates with high plasma protein binding.

The HPLC technique has great potential to determine compound distribution properties through retention time measurements. Various stationary phases can be used, such as C18, immobilized artificial membrane (IAM), human serum albumin (HSA), alpha-acid glycoprotein (AGP) and others.
